Application of superabsorbents in soil and its influence on plant growth

This thesis is focused on description of superabsorbents with controlled released of nutrients, specifically macronutrients nitrogen, phosphorus and kalium. Superabsorbents are polymers, mostly acrylic acid polymers, that can absorb huge amount of water without dissolving in it. In its structure they can hold water and thanks to this they are very perspective materials for agricultural usage. Experimental part was focused on growing corn with use of superabsorbents and control samples without use of superabsorbents. Plants were growth for 3 time periods, specifically 46, 60 and 74 days. During the experiments, plats were growth without water stress and in controlled conditions in growing box under UV lamp that simulated day and night by turning on and off after 12 hours. At the end, an image analysis was performed in HARFA program, from which we got information about branching of the roots. Results were supplemented by soil and plants analysis.
Influence of makroelements from food on DNA and epigenetic profile

The macroelements contained in food have an important function for the human body. They are involved in several of biochemical reactions in the body and their abundance can prevent serious diseases. The theoretical part of the bachelor thesis describes the function of minerals in the human body, the function of DNA and epigenetic mechanisms such as DNA methylation or histone modification. The influence of nutrition and function of selected macroelements – sodium, magnesium, calcium, potassium on epigenetic modifications and on the stability of G-quadruplexes was described. The aim of the experimental part of this work was to study the effect of these substances on DNA structures in vitro and to prepare them experimentally for in vivo studies.
Design of WWTP at locality with extreme demands on quality of treated wastewater

This diploma thesis deals with the wastewater treatment technologies by means of which high quality of the treated wastewater is attained, especially with regard to removing nutrients. The introduction of the thesis contains, among others, descriptions of the reserves in the Czech Republic where it is extremely important to insist on the highest quality of the discharged wastewaters. Then follows a summary of the quality requirements on the discharged wastewaters as stipulated in the legislation. Furthermore, I analyse the impact of the nutrients – nitrogen and phosphorus – on the environment and the principles of their removal from the wastewaters. In the next section I concentrate on the technological processes by means of which the required quality of the wastewater is achieved. The conclusion includes a list of technologies used as the tertiary wastewater treatments with emphasis on membrane technologies. The second part of my thesis is a study of a project of a wastewater treatment plant (WWTP) with membrane technology for the villages of Sloup and Šošůvka located within the protected landscape area of Moravian Karst.
Use of combination of bacteria and algae for wastewater treatment

The aim of this bachelor's thesis is to introduce the reader to the ever improving methods of biological wastewater treatment with the use of bacteria, algae and combination of both. Further it elaborates on the promising ways of utilizing biomass gained from these treatments in various sectors, such as energetics, chemistry, pharmacy and agriculture. The second part of the thesis offers a techno-economic study of wastewater treatment using combination of bacteria and algae in a specifically assigned area.
Balance of energy, water and nutrients in the aquaponic cycle

Předložená diplomová práce byla zpracována s cílem vytvořit přehled poznatků v oblasti akvaponické potravinové produkce. Informace získané během tvorby tohoto přehledu pak měly vést, v kombinaci s daty získanými z funkčního provozu, k vytvoření matematického modelu akvaponického cyklu. Na akvaponické farmě provozované společností Flenexa plus s.r.o., která byla zdrojem potřebných procesních dat, měla být dále zpracována a vyhodnocena bilance energie a vody. Nakonec měla být v průběhu práce posouzena možnost implementace mikrořasového fotobioreaktoru do akvaponického cyklu. Úvod práce představuje motivaci vedoucí k potřebě inovovat dnešní potravinovou produkci. Kriticky jsou zhodnoceny predikce vývoje lidské populace, a to pak hlavně z pohledu dopadu, který by tento růst měl na zemědělskou produkci. Současná situace se na základě získaných poznatků ukazuje jako neudržitelná, primárně pak v oblastech vodohospodářství a energetické spotřeby. Následně je jako možné řešení vedoucí ke zlepšení udržitelnosti potravinové produkce zkoumána akvaponie. Akvaponie je definována a její jednotlivé komponenty jsou představeny z hlediska mechanismu jejich fungování a z pohledu jejich návrhu. Mezi popsané oblasti patří například principy tzv. coupled a decoupled akvaponie a popis možných typů hydroponického komponentu. V této části práce je pozornost věnována také představení cyklů jednotlivých živin v rámci akvaponie. Následující a poslední teoretická část práce je pak věnována mikrořasovému fotobioreaktoru. Jsou zde popsány mechanismy, jak motivující, tak odrazující od zakomponování bioreaktoru do akvaponie. V oblasti výhod se jedná hlavně o jeho roli ve stabilizaci pH a spotřebě toxikého amoniaku. Na druhou stranu jeho ekonomické dopady na profitabilitu akvaponie jsou velmi proměnlivé v závislosti na způsobu implementace. Samotný mikrořasový fotobioreaktor je pak v práci detailněji představen. Jednotlivé procesní ukazatele ovlivňující růst řas jsou rozebrány, a to společně s jednotlivými typy fotobioreaktoru, metodami sklizně a využitími pro vyprodukované mikrořasy. Na základě poznatků schromážděných v této práci pak lze jako nejvhodnější k implementaci do akvaponie doporučit hybridní fotobioreaktory, u kterých je většina osvětlení zajištěna v podobě slunečního svitu. Samotná experimentální část práce pak začíná popisem zkoumaného provozu společnosti Flenexa plus s.r.o. z pohledu aplikovaného akvaponického procesu. Jednotka podrobená měření byla provozně stabilní a využívala implementace hydroponického komponentu typu Deep Water Culture (DWC). Spolu s detailním popisem celého provozu jsou poskytnuty a vyhodnoceny vypracované bilance vody a energií. Pozornost je pak přesunuta k matematickým modelům vypracovaným a ověřeným na základě dat a poznatků shromážděných z provozu společnosti Felenexa plus s.r.o. Logika a algoritmy, na kterých jsou oba modely postaveny, jsou v této části vysvětleny a diskutovány společně s hlavními funkcemi a schopnostmi obou modelů. První, primárně statistický model je představen jako nástroj pro použití při uvádění akvaponie do provozu. Druhý, fyzikální model pak v uživatelsky přívětivém formátu představuje základ pro model řízení akvaponické farmy s mikrořasovým fotobioreaktorem. V neposlední řadě jsou nastíněny také cesty možného budoucího vývoje pro oba vytvořené modely. Práce je následně završena shrnutím a diskusí nad poznatky a výstupy získanými během celého tvůrčího procesu.
Determination of the nutritional properties of paprika of different geographic origin

This master thesis deals with the analysis of nutritional properties of paprika condiment with different geographic origin. The theoretical part contains botanical description of red pepper (Capsicum annuum L.), its chemical composition, process of production of paprika and then they are described analytical methods suitable for determination of fundamental nutrional properties. Experimental part is focused on the preparation of samples and their analysis. In total, 11 samples were selected for the experiment, 5 of them had a protected designation of origin. These samples were from Slovakia, Hungary, Bulgary, Romania, Turkey and Spain. The Soxhlet extraction was used for the analysis of the fat content, Kjeldahl method was used to determine gross protein. High-performance liquid chromatography (HPLC) was chosen to determine carbohydrates and capsaicin. Induction-coupled plasma emission spectroscopy (ICP-OES) was used for elemental analysis. The results were compared with each other and with available databases and literature. For comparing and finding certain connections, the principal component analysis (PCA) was selected.
Superabsorbents with controlled release of nutrients

This thesis is focused on the study of superabsorbent polymers and their ability of control release of nutrients. These polymers can absorb huge amount of water, up to hundredth times more than the original weight is. In combination with nutrients, especially nitrogen, phosphorus, potassium and lignohumate, they make effective fertilizer. Due to this fact, the possibility of use in agriculture is increasing. Therefore, experiments that are researching the ability of containing the water were performed, the swelling capacity was measured. Experiments that are related to control release of nutrients were performed next. Therefore, this thesis is focused on the study of superabsorbent polymer characterisation, that can be used in agriculture.
Changes in rainfall-runoff regime and biogeochemistry in headwater areas

Headwater areas are a very important current or potential drinking water source. Their ecological value for the whole ecosystem is equally important. Headwater areas are sites with minimal anthropogenic intervention and usually the least polluted water sources. Changes in climate and rainfall-runoff (R-R) regime together with water quality in these areas affect the status and availability of water further downstream significantly. It is also one of the most sensitive areas and is an early indicator of ongoing changes. This thesis focuses on the main issues that are already ongoing and are affecting the availability of drinking water significantly. Some of these are trends in R-R regime in the context of the climate change and glacier retreat in headwater areas. As a very good example of a mid-latitude river with a complex rainfall-runoff regime, where a large impact of regional climate warming and glacier retreat are strongly manifested, The Athabasca River Basin was selected for this study. Study of the limnological components of the ecosytem of Lake Medard in 2022

The report summarizes the results of research into the limnological and biological components of the ecosystem of the newly created Medard post-mining lake. The aim of the presented report is to summarize the results achieved for the year 2022 and recommendations for further monitoring. The year 2022 was the fifth year after the lake was completely filled, when water from the Ohře River was no longer added. Only small tributaries flowed into the lake from its own catchment area, and at the same time, water from the lake drained through seeps, which are apparently drained into the nearby rivers, the Ohře and the Svatava. The lake ecosystem reached the final state of its initial phase in terms of hydrological regime and nutrient load. This condition is characterized by very little water exchange and a low nutrient level with low productivity of plankton communities. There is the development of submerged macrovegetation in littoral zone and a specific seasonal change in chemistry and biological parameters. However, the aquatic ecosystem is still far from stabilized and the biological communities of the littoral zone (littoral) and open water (pelagial), including the fish community, still show significant interannual dynamics.

The bachelor thesis deals with the issue of nutrition of pregnant mares and is processed in the form of a literature search. At the beginning of the thesis there is an introduction to the subject, which is devoted to the characteristics of the equine sexual cycle and foaling. The aspects that influence them are described, as well as the reproductive and digestive organs, the development of the foal in the mother’s body and a description of the body score condition of the horse. Subsequently, two chapters are focused to the nutritional requirements of mares in early and late foaling and the differences between them. The last chapter describes specific types of feeds and their suitability for broodmares. Selected complete feed mixtures – granules and mueslis – are also summarised, with a description of their composition, recommended dosage and in tables is shown the content of the most important essential nutrients – digestible energy, starch, nitrogenous substances, fat, fibre, calcium, phosphorus, vitamins A and E and lysine.
